I wouldn't know how it's different from Facebook or Twitter, also, as I don't have a Facebook or a Twitter, but for me the appeal is that
I can say whatever I like about anything I'd like, and the people who are interested will reply, while the people are not have no obligation
as opposed to real life conversations, in which I try to muddle about finding something in common and never leave Awkward Land.
But isn't the 'muting' going to be something you'll have to continuously do?

no, that particular factor isn't unique to plurk. it's more of an internet thing than anything else.
plurk is like a mix of twitter and lj.
but plurk is nice because you can bring up absolutely useless things that you want to talk about anyhow, like...
it's easier to navigate than twitter, and each plurk functions like a little chatroom
you could make a plurk about stubbing your toe or such, but in a forum that sort of post would be considered an unnecessary waste of space
but without the exclusion that can come from walking into a chat too late and not know the context. people can join in on a conversation
you could mention it in a chat, but chats are single linear conversations
